Am isohybrid file can be used as both harddisk and CD-image. This
behaviour can be tested in Bochs emulator (and likely QEMU as well) by
specifying the isohybrid file first as the Virtual CD-ROM device, and at
2nd attempt as harddisk. Both should work (at least for me, using DOS).
Once a virtual system works, you can try some USB stick with Syslinux
and memdisk on it, and load your ISOHYBRID file by MEMDISK in both modes
(memdisk hybrid.iso append harddisk ; memdisk hybrid.iso append iso).
Finally, use something like DD or PowerISO to raw-write the Isohybrid
file to an empty USB stick and then boot from it. With a bit of luck you
can partition the remainder of the USB stick as well if so desired
(that's the aim of having a hybrid file that has a filesize of an exact
amount of MBs).
Be aware Windows might not like removable disks with multiple
partitions, and it will request to repartition/format the device.
As you're experiencing, booting an operating system present beyond the
first (hybrid/ISO9660) partition can be slightly challenging.
